Goniothalamin: Isolation and In vitro anti plasmodial and Hemolysis Study

Abstract

Phytochemical studies of theGonothalamus scortheciniiKing have resulted in the isolation and characterization of many compounds with a variety of biological properties. Goniothalamin is one of the natural styryl lactones isolated from the plant. It has been reported to have weak antibacteria (gram positive and gram negative bacteria) and significant antifungal activity in vitro. It is insecticidal and widely reported to have cytotoxic properties in a variety of cell lines. The present study demonstrates the isolation of Goniothalamine and to investigate the in vitro biological activity of goniothalamine forits potential as anti malaria. Anti plasmodia testing, cytotoxicity to normal cell line, its effect (in vitro insult) on erythrocytes and its possible mode of action were studied in vitro. The anti plasmodia activity was carried out using the parasite lactate dehydrogenase (pLDH) assay. Goniothalamin was tested over a series of concentration from the highest 64 μg/ml tothe lowest 0.03 μg/ml. The reaction was incubated for 72 hours, followed by determination of pLDH concentration by a coenzyme. The conversion of tetrazolium to blue formazan was an indicator for the test system and absorbance was measured by ELISA reader. The cytotoxicity test was performed using the MTT assay on a series of cell lines including MDBK cells. The hemolysis assay was carried out to determine the effect of Goniothalamine on erythrocytes. A series of Goniothalamine concentration and erythrocytes suspension (10% in PBS) were reacted in a ratio of 1:5 and incubated overa series of incubation period. The percentage hemolysis were determined. The study showed that in vitro bological activity of Goniothalamin toP. falciparumresistant strain is good with an IC50 of 8.9 μg/ml. The IC50 values to MDBK was 20.00 ± 12.72, indicating a selectivity index of more than 1. The activity to determine the in vitro insult to erythrocytes demonstrated a positive indication.
