Comparing The Cytotoxic Effect of Taxol in The Presence of Different Solubilizing Agents

Abstract

Taxol, a poor soluble diterpen compound, isolated from the bark of Taxus brevifolia (Pacific Yew), possesses high cytotoxicity and strong antitumor activity. A major barrier to evaluate the antitumor response of many anticancer compounds in mammalian cells models can be attributed to the poor water solubility of many of these compounds. Utilization of co­solvents or solubilizing agents is an effective approach to overcoming this problem. The effect of various solubilizing agents in overall cytotoxicity of taxol was evaluated. KB cells were cultivated in CS-MEM medium, seeded in 96 well plates and incubated at 370C in a humidified, 5% CO2atmosphere. Results showed that comparing to water, the cell toxicity effect of taxol in the presence of nicotinamide, liposomes, and PEG decreased while present of bile salts, ethanol and  cremophorEL  increased  the  cytotoxic effect of taxol (P < 0.05). HPβCD and γCD did not change the cytotoxicity of taxol (P < 0.05). In conclusion, it should be considered that solubilizing agents and additives potentially have some intrinsic cytotoxicity which may change the cytotoxic activity as well as other physicopharmaceutical, pharmacokinetic and pharmacodynamic properties of the active natural products.
