Abstract

Traditional herbal medicines are chemically very complex pharmaceutical preparations. Very often, the observed in vitro or in vivo activity cannot be related to a single constituent, but is a complex function of numerous constituents. Yet, herbal medicine is often standardized using a single or just a few constituents, and thus individual preparations may be highly variable with respect to other constituents that directly or indirectly affect pharmacological effect. Description of composition of herbal medicine can be conveniently done using metabolomics tools such as multivariate analysis of NMR data that contain information about all constituents present above detection limit of the method. Individual components of the extracts, including yet unknown constituents as well as constituents accounting for variability of a preparation, can be conveniently identified with hyphenated NMR methods in the microgram concentration domain. The obtained information is of pertinent interest in relation to observed or anticipated pharmacological activity. In this lecture, examples of application of NMR-based metabolomics and hyphenated NMR methods (in particular HPLC-SPE-NMR) for characterization and analysis of medicinal plants will be discussed. The examples include plant extracts as well as commercial herbal preparations (Hypericum perforatum,Ginkgo biloba,Harpagophytum procumbens,Echinacea purpurea,Warburgia salutaris, andKanahia laniflora).
