Abstract

The crude methanol extract of the leaves of Garcinia nervosa var. pubescens, King (Guttiferae) was investigated for its inhibitory effect on platelet-activating factor (PAF) receptor binding using rabbit platelets at concentration of 18.2µg/ml. The crude extract showed significant inhibitory effect of more then 60 % inhibition. The crude extract was then fractionated into ethyl acetate, butanol and methanol, successively. The ethyl acetate fraction gave the highest inhibitory activity with inhibition of 78.0 %, while the butanol and methanol fractions exhibited 47.8 and 36.2 % inhibition, respectively. Vacuum liquid chromatography (VLC) of the ethyl acetate fraction afforded seven fractions, N(I) to N(VII). Fractions N(V) and N(VI) which showed significant inhibitory effects of 72.6 and 66.7 % respectively, were rechromatographed on silica gel to give several active fractions. Further purification of the active fractions yielded four compounds (1, 2, 3 and 4) with PAF antagonist activity of 29.5, 35.7, 70.0 and 73.9 %, respectively.
