Effect of Andrographis paniculata on Isolated Rat Hearts: A study from crude to isolated compounds

Abstract

The cardiovascular activity of the Andrographis paniculata Nees (AP) extracts (aerial part) were screened for effects in isolated perfused rat hearts. Three different AP extracts, i.e. hexane, dichloromethane and methanol were prepared and tested for the activity. All, except the methanol extract, dilated coronary vessels. The heart rate was reduced by the dichloromethane extract. The methanol extract was found to be inactive. Further isolation and purification was carried out on the dichloromethane extract. Five andrographolide series were isolated and identified as andrographolide 1, 14-deoxy-11,12-didehydroandrographolide 2, 14-deoxyandrographolide 3, 14-deoxyandrographolide 4 and neoandrographolide 5. The structure identification and elucidation was done using normal spectroscopic technique (NMR, IR, UV and MS) and by comparison of the spectral data with the literature values. These studies led to the discovery of compounds responsible for coronary vasodilation in rat hearts, i.e. 14-deoxy-11,12-didehydroandrographolide 2 and 14-deoxyandrographolide 4. The difference of effects on the isolated rat hearts between the crude extract (dichloromethane) and isolated compounds suggested synergistic or complementary activity between the compounds.
