Abstract

The Zingiberaceae family is distributed worldwide and is known not only as an essential food but has been used throughout history for their medicinal effects. Traditionally, Zingiberaceae is used for the treatment of abdominal bloating, coughing, vomiting, diarrhea and rheumatism. In the present study, three species of Zingiberaceae namely Zingiber officinale (halia), Curcuma xanthorrhiza (temu lawak) and Curcuma longa (kunyit) were screened for cytotoxic activity against HCT-116 cells by the in vitro neutral red cytotoxicity assay. The HCT-116 cells, a colon cancer derived cell line, grown in 10% McCoy’s were treated with crude petroleum ether, chloroform and methanol extracts of the plants at concentrations of 0, 1, 10, 25, 50 and 100 µg/ml for 72 hrs and then incubated with Neutral Red for 3 hrs. Absorbance readings were taken at 540 nm and the IC50 values determined.  The cytotoxic effect was dose-dependent, i.e. as the concentrations of extracts increased the percentage of inhibition increased. Five extracts showed IC50 values less than 20 µg/ml. Crude petroleum ether and chloroform extracts of C. xanthorriza exhibited the most potent cytotoxic activity with an IC50 values of 16.8 µg/ml and 15.0 µg/ml, respectively. The IC50 values for crude petroleum ether, chloroform and methanol extracts of C.longa were 17.1 µg/ml, 6.1 µg/ml and 16.7 µg/ml, respectively. Overall, the chloroform extracts of selected Zingiberaceae showed greater cytotoxic activity against HCT-116 cells. The results suggested that the plants with high cytotoxic activity could be studied further for their anticancer properties.
