Abstract

Ficus deltoidea Jack (Moraceae), locally known as Mas cotek or Serapat angin, has been widely utilized in Malaysia for various medicinal purposes including the treatment of diabetes mellitus. An increase in the popularity of its use entails scientific evaluation on its safety. This study evaluated both acute and sub-chronic toxicity profiles of the leaf methanol extract of F. deltoidea in streptozotocin-induced diabetic rats. The rats (n=5/group) were subjected to acclimatization for 7 days and fasted for 15 h prior to experiments. The acute toxicity evaluation proceeded through administering orally to the rats the methanol extract, ranging from 1600 to 6400 mg/kg body weight in 10% w/v Tween 80 solution. The extract-fed rats were subjected to macroscopic observation at 6, 12, 24, 48 and 72 h for overt signs of acute toxicity and death. The sub-chronic toxicity test was conducted by administering orally 200 mg methanol extract/kg body weight to rats once daily for 28 days. These rats were examined for symptoms of toxicity, food and water consumption pattern, body weight and urination and defecation profiles weekly. Their biochemical and histological profiles of Liver and kidney at the end of 28 days were also studied. There were no overt signs and death among control and extract-fed rats in acute toxicity study. In the sub-chronic study, administration of rats with methanol extract did not bring about marked changes in the behaviour, body weight, food and water intake, urination and defecation pattern, biochemical parameters (urea, bilirubin, creatinine, alanine aminotransferase and aspartate aminotransferase) as well as histological profiles of liver and kidney when compared to the control group. The methanol extract of F. deltoidea could be considered at least as non toxic for consumption by the diabetic rats via the oral route.
