In Iranian traditional medicine Vitex agnus-castus leaf and fruit extracts are used for milk increasing. In present research the effects of Vitex agnus-castus leaf and fruit extracts on serum concentration of prolactin hormone has been investigated. Selective dosages of Vitex agnus-castus were orally administrated to the female rats during proestrus stage of estrus cycle and lactation. In the control group of rats the distilled water was administrated instead of Vitex agnus-castus extract. In order to blood sampling from each rat, at each stage (procestrus, and 3rd day of lactation) the animal was anaesthetized by inhalation of ether and the fresh blood sample was taken off from its heart. The serum was separated from each sample by centrifugation. These serums were used for measurement of prolactin hormone by RIA (Radio Immono Assay) method. The data were analysed by ANOVA, MANOVA and LSD test at P < 0.05. It is concluded that oral administration of 70 mg/kg/day of Vitex agnus-castus extract in lactation stages, significantly increase serum prolactin when compared to the control group of rats. The effect of Vitex agnus-castus extract on the difference of sampling weight and stating weight (before administration) was not significant in comparison to the control group rats.
