XanthorrhizoI is a sesquiterpenoid Isolated from the rhizome of Curcuma xanthorrhiza which known as ‘temu lawak’ derived from the family Zingiberaceae. The purpose of this study is to invistigate the anti proliferative effect of xanthorrhizol and the mode of cell death towards human malignant melanoma cell line, HM3K0. The data obtained showed that xanthorrhizol displayed anti-proliferative effect towards HM3KO with EC50 of 4.01±0.05 µg/ml. Howewer, xanthorrhizol showed a non-cytoselective characteristic towards normal cell line CCD1114sk (fibroblast). The extent of DNA fragmentation was also was also evaluated using Tdt-mediated dUTP nick end Iabellling (TUNEL) assay. XanthorrhizoI was found to induce apoptosis in HM3KO cells with morphological changes such as fragmentation of DNA, cell shrinkage, cytopiasmic and chromatin condensation all being typical of apoptosis. The apoptotic index (AI) showed an increase after 24, 48 and 72 h exposure at 30, 69 and 94%, respectively. These results concluded that xanthorrhizol exerted anti-proliferative effects on HM3KO cells through the activation of apoptosis.
