Due to significant expansion on the use of medicinal plants as alternative medicine, it is very important to have good standard methods for quality control in assessing the multi-component mixture of compounds in medicinal plants in order to guarantee consistencies and authenticities. Proper authentication of the raw materials is of critical importance to the safety and efficacy of the resulting products. Products using plants of mistaken identity could result in health hazard for the consumer. The routine botanical species identification represents an unfeasible approach because herbal manufacturers often buy the pre-processed raw materials. Thus chemical fingerprint technique is a necessity in the quality monitoring of the raw material. The Multi-steps infrared (IR) macro-fingerprinting provided a new and powerful non-destructive approach for rapid discrimination of plant Identities. Besides, Multi-steps IR macro-fingerprinting technique, analytical separation technique such as high performance liquid chromatography (HPLC) also routinely used for this purpose. Both techniques are able to confirm the authenticity of plant material which is indicative of the consistency of raw material and herbal product quality. Labisia pumila (Blume) Fern.-Vill. or locally known as Kacip Fatimah is chosen not only for its economic importance but also because there are three varieties that are morphologically very similar. The industry always uses the name ‘Kacip Fatimah’ and very likely a mixture of these three varieties. This species, including these three varieties have been used by women in Malaysia for many years to induce and facilitate childbirth, as well as a treatment for menstrual irregularities and post-partum. The herb belongs to a small genus of slightly woody plants of the family Myrsinaceae and is found In Indochina as well as throughout Malaysia. It grows wild in the Malaysian rainforests at about 80-100 meters above sea level. The three varieties are differentiated base on the characters of the petioles. The differentiation can be difficult especially for layman due to similarity in the overall. Each variety commands a different usage and thus, it is Important to ensure that the right variety is used in each case. This paper reports on the chemical characterization of three varieties of L. pumila using the approach of multi- step IR macro-fingerprinting arid HPLC techniques. This study also conducted by Incorporating appropriate chemometrics method (Principal Component Analysis, PCA) as tools for extracting relevant chemical information from the IR data. Furthermore, the visual and colourful 2D-IR spectra obtained by applying thermal perturbation enhanced the discrimination among the L. pumila varieties more effectively. Consequently, the macroscopic fingerprint characters of FT-IR and 2D-IR spectrum complemented with HPLC chromatograms indicated many holistic variation rules of chemical constituents of the different varieties of L. pumila. This approach can be used to demonstrate a powerful, useful and non-destructive approach for the authentication of L. pumila.
