Cytotoxic Effect of Gracilaria changii on Ovarian Cancer Cells

Abstract

This study reports the isolation and identification of a cytotoxic compound from Gracilaria changii collected from Morib, Selangor. The extracts were subjected to thin layer and column chromatography to separate and isolate the bioactive compound. The structure of the compound was elucidated on the basis of comprehensive spectral analyses (IH, 13C, COSY, HMQC and HMBC NMR) as well as GC-MS analysis. The cytotoxic activity against the CaOV3 cell line (anti-ovarian cancer) was tested on the methanol , butanol, diethyl ether extracts of the plant and the pure isolate (GCM 1). This cytotoxocity test was assessed by the MTT assay.  The methanol extract showed negative result (IC50>30μg/ml) whereas the butanol extract and the diethyl ether extract demonstrated cytotoxicity at IC50 22 μg/ml and 10 μg/ml.  The pure compound (GCM 1) was active against the CaOV3 cell line with the IC50 value of  < 10 μg/ml.
