Effect of Dietary Tocotrienol and Tocopherol Supplementation On Parcetamol Toxicity In Mice

Abstract

Three groups of mice were given tocotrienol supplemented diets (30, 60 and 150 mglkg pellet), one group was given tocopherol supplemented diet (34 mglkg pellet) and the control group received the pellets without additional Vit. E supplementation, all for 8 weeks. The dietary treatments did not significantly affect the 24 hour LD50 values for Paracetamol. Plasma alanine aminotransferase (ALT) activities measured four hours after a single dose of PCM (250mglkg), i.p. increased significantly in the group that received tocopherol compared to the groups receiving tocotrienol but not to the control group. In addition some mortalities were noted at this time point which was highest in the control group and least in the tocotrienol groups. This result shows that tocotrienol appears to provide a better protection PCM toxicity in mice compared to tocopherol.
