Coleus species are widely used in the region of tropical countries such as Vietnam, India and Indonesia as remedies for heart diseases, abdominal colic, respiratory disorders, painful maturation and certain central nervous system diseases. In Malaysia. Coleus spp. are locally known as ‘Ati-ati’ plant and used primarily as decoration houseplant. The use of Coleus spp. as remedies in Malaysia was unknown. The study was aimed to determine total antioxidants content in three different Coleus spp., namely, C. amboinicus Lour. (CAL), C. blumei (red hybrid) (CBR) and C. blumei (mixed colour hybrid) (CBMC). The quantities of total antioxidants in different parts (leaves, petioles and stems) of the plants were evaluated. Fresh CAL samples were randomly collected from Jakarta, Indonesia and samples of CBR and CBMC were randomly collected from Kuantan, Malaysia. All samples were freeze-dried, ground and extracted using 60% methanol. The antioxidant content of the extracted samples were measured using the scavenging activity of the stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical (DPPH radical-scavenging assay). Ascorbic acid was used as positive control and reaction mixtures contained an ethanol solution of 200 IM DPPH (100 µl) and two fold serial dilutions of sample (dissolved in 100 µl ethanol, with amounts of sample ranging from 2 to 1000 lg/mI). Absorbance at 517 nm was determined after 30 mm and percent inhibition activity was then calculated. Results showed that the total antioxidant contents in CBR and CBMC were significantly higher (p<O.O5) than in CAL but there was no significant difference (p>O.05) in total antioxidant content in CBR and CBMC. All different parts of CAL exhibited similar (p>0.05) amount of total antioxidants content. In CBR and CBMC, the leaves showed the highest (p<O.O5) total antioxidant content followed by petioles and stems. Coleus blumel showed greater amount of total antioxidant content compared to C. amboinicus. The study showed the possibility of Coleus spp. from Malaysia for natural functional food.
